Parmi eux, le SRP de type II (Majewski ; MIM 263520) se caract\u00e9rise par des tibias courts et ovo\u00efdes ou une ag\u00e9n\u00e9sie tibiale et est radiographiquement tr\u00e8s proche du SRP de type IV (Beemer-Langer ; MIM 269860) qui se distingue par des radius et des cubitus arrondis et des tibias relativement bien tubul\u00e9s. Des mutations NEK1 ont \u00e9t\u00e9 r\u00e9cemment identifi\u00e9es dans le SRP de type II. La double h\u00e9t\u00e9rozygotie pour les mutations dans les deux NEK1 et DYNC2H1 dans un cas de SRP de type II supporte une possible h\u00e9r\u00e9dit\u00e9 dig\u00e9nique diall\u00e9lique.<\/p>\n\n\n\n<p><strong>M\u00e9thodes :&nbsp;<\/strong>L'objectif de cette \u00e9tude \u00e9tait de d\u00e9pister DYNC2H1 et NEK1 dans 13 cas de SRP de type II et dans sept cas de SRP de type IV. Il n'a pas \u00e9t\u00e9 possible de d\u00e9pister DYNC2H1 chez deux patients en raison d'une quantit\u00e9 insuffisante d'ADN.<\/p>\n\n\n\n<p><strong>R\u00e9sultats :&nbsp;<\/strong>L'\u00e9tude a identifi\u00e9 des mutations homozygotes de NEK1 dans 5\/13 SRP de type II et des mutations h\u00e9t\u00e9rozygotes compos\u00e9es de DYNC2H1 dans 4\/12 cas. Enfin, NEK1 et DYNC2H1 ont \u00e9t\u00e9 exclues dans 3\/12 cas de SRP de type II et dans tous les cas de SRP de type IV. La principale diff\u00e9rence entre le groupe SRP de type II \u00e0 mutation positive et le groupe SRP de type II \u00e0 mutation n\u00e9gative \u00e9tait la pr\u00e9sence d'holoprosenc\u00e9phalie et de polymycrogyrie dans le groupe \u00e0 mutation n\u00e9gative.<\/p>\n\n\n\n<p><strong>Conclusion :&nbsp;<\/strong>Cette \u00e9tude confirme que NEK1 est un g\u00e8ne responsable du SRP de type II mais signale \u00e9galement des mutations dans DYNC2H1, \u00e9largissant ainsi le spectre ph\u00e9notypique des mutations de DYNC2H1.
